I installed my Splunk demo on a Windows 64-bit VM with 2 physical processors assigned to it, then the VM admins gave me 2 additional processors, do I need to do anything to the config for Splunk to optimize the use of them since they did not exist at install time?

No, you don't need to do anything for Splunk to take advantage of them. Once you reboot, you should be all set, Splunk will take care of it for you.
