We plan to update our older Splunk Enterprise Environment (5.xx) for which we have Perpetual Enterprise Licenses to an up-to-date Enterprise Version. Do we need to buy/reactivate the Annual Enterprise Support Licenses to be allowed to do so?

The short answer is No. Your license is only associated with your indexing volume, not your Splunk Enterprise version. You can always upgrade to the latest Splunk Enterprise version without refreshing your license. For upgrade instructions, please refer to the documentation here:

Please note that a new "no-enforcement" license type that does not block search after a license has been in violation is available, for Splunk 6.5.0 and later. For details, see the documentation here:

i dont think upgrade has anything to do with the license.
license only counts for indexing ratio. I will advise to co reactivate your support regardless. (and specially since it will be a major upgrade and you might need help).
