From Splunk DB Connect documentation:
Run : splunk cmd python $splunk_home/etc/apps/dbx/bin/reload.py databases
It can reload database.conf without restart Splunk.
I need to script schedule update/modify DB input conf.
Any command can run like as above, reload input.conf without restarting Splunk.
But, I found "Note: Modifying inputs.conf file stanzas outside of the DB Connect app, such as in the search app or manager context, is not supported."
Is it possible to reload inputs.conf without restarting the Splunk Service?

If you are using a unix host, try the following from /opt/splunk/bin:
splunk _internal call /services/apps/local/dbx/_reload -auth admin:$ADMINPWD
It is important to note that although some components of the app can be reloaded in this manner (databases.conf for password encryption), other components (like the java bridge server) will require a restart as per the output message from the command above:
Restart required by:
ui-prefs, authorize, java, web, ui-prefs

For DB Connect v2, I had to use this command (sudo as splunk user):
./splunk _internal call /services/apps/local/splunk_app_db_connect/_reload -auth admin:$ADMINPWD
